RESUMEN Debaryomyces hansenii es la especie de levadura más abundante en embutidos curados fermentados, y se ha asociado a los cambios proteolíticos que acontecen en dichos productos, y que van a ser determinantes en el desarrollo del sabor y del aroma de los mismos. Sin embargo, no existen estudios bioquímicos en los que se describan la composición y posibles funciones del sistema proteolítico de D. hansenii. Por ello, el objetivo de la presente tesis fue el estudio del sistema proteolítico de D. hansenii, mediante la purificación y la caracterización de los enzimas más importante, así como de los factores que afectan a su síntesis. En primer lugar se seleccionó la cepa D. hansenii CECT 12487 por su mayor actividad proteolítica. Esta cepa fue capaz de hidrolizar las proteínas sarcoplásmicas de músculo porcino generando péptidos y aminoácidos libres de importancia para el sabor, y como precursores de compuestos aromáticos. La inoculación de cultivos viables o extractos celulares permitió modular cualitativa y cuantitativamente los productos resultantes. Los estudios de purificación y caracterización revelaron que el sistema proteolítico de D. hansenii está constituido, al menos, por dos aminopeptidasas: una prolín aminopeptidasa (PAP) y una arginín aminopeptidasa (AAP); y dos endoproteasas: la proteasa A (PrA) y la proteasa B (PrB). Las principales características de las enzimas purificadas son las siguientes: la prolín aminopeptidasa (PAP) de D. hansenii es una cisteín peptidasa de 370 kDa, compuesta por subunidades de 53.5 kDa, activa en un amplio intervalo de pH (5.0-9.5), con un óptimo de actividad a pH 7.5 y 45 ºC, y que libera exclusivamente prolina del extremo amino de las proteínas y de los péptidos. La PAP no había sido descrita previamente en levaduras. La arginín aminopeptidasa (AAP) de D. hansenii es una metalo peptidasa de 101 kDa y estructura dimérica, con pH de actuación neutro-básico (6.0-9.0) y óptimo de actividad a pH 7.0 y 37 ºC. La AAP de D. hansenii es homóloga a la ApY, también denominada ApCo, descrita en Saccharomyces cerevisiae, activada por cobalto y con preferencia por aminoácidos básicos (arginina y lisina). La proteasa B (PrB) de D. hansenii es una serín proteinasa de 30 kDa y con un pH de actuación neutro-básico (6.0-12.0) y óptimo de actividad a pH 8.0 y 37 ºC. Por sus propiedades bioquímicas, la PrB de D. hansenii es homóloga a la PrB de S. cerevisiae. La producción de las enzimas proteolíticas estudiadas, PAP, AAP, PrB y PrA, depende de las fuentes de nitrógeno y carbono utilizadas en el medio de cultivo y de la fase de crecimiento. Las actividades específicas aumentan como respuesta a la presencia de fuentes pobres o complejas de nitrógeno y carbono, y a medida que el crecimiento celular avanza hacia la fase estacionaria. La obtención de cultivos o extractos de D. hansenii utilizando urea como fuente de nitrógeno, así como la prolongación del tiempo de incubación para alcanzar niveles máximos de actividad total, podría constituir una buena alternativa para su incorporación a la masa cárnica, y así lograr la mejora del proceso de maduración de embutidos curados. __________________________________________________________________________________________________

Proteolysis is one of the key factors in the flavour development of dry-cured fermented sausages. On the other hand, Debaryomyces hansenii, is the more abundant yeast specie in fermented sausage, and it has been associated with the proteolytic changes that take place in cured products. However, there are very few biochemical studies related to the characteristics and possible functions of the proteolytic system of D. hansenii. Therefore, the main objective of the present Thesis was to study the proteolytic system of D. hansenii, by means of the purification and characterisation of the most important enzymes as well as the factors affecting to its synthesis. Firstly, the strain D. hansenii CECT 12487 was selected by its highest proteolytic activity. This strain was capable to hydrolyse the muscle sarcoplasmic proteins generating peptides and free amino acids of importance for the taste, and as precursors of aromatic compounds. The inoculation of viable cells or cell extracts allows to modulate qualitative and quantitatively the resulting products. The purification and characterisation studies revealed that the proteolytic system from D. hansenii, it is constituted at least, for two aminopeptidases, a prolyl aminopeptidase (PAP) and an arginyl aminopeptidase (AAP), and two endoproteases, the protease A (PrA) and the protease B (PrB). The characterisation of these enzymes and its possible influence during the curing of meat products it is described in the corresponding sections of the PhD Thesis. The specific proteolytic activities of the studied enzymes, PAP, AAP, PrB and PrA, were increased in poor or complex nitrogen and carbon sources, and at the time, the cellular growth advances toward the stationary phase. The production of cultures or cell extracts of D. hansenii using urea as nitrogen source, as well as long times of harvest, permit to reach maximum levels of total proteolytic activity. And therefore, it could constitute a good alternative for its incorporation to the meat formulation, and to improve the ripening of cured sausages.
